Tissue decalcification:

Formalin solution

  1. 10% neutral buffered
  2. 24 hours


  1. 10% pH 7.2 (e.g. in 1% PBS)
  2. 72 hours

Maintain the specimens in containers during these steps and gently shake on a slow horizontal orbital lab shaker. Set to very slow low speed, but that is probably not essential. The timing stated is for a core biopsy, if on the other hand it is a large surgically excised chunk of tissue you will want to double the time in EDTA and change the solution once half way through the time.
